Simply trying it would have anwsered your question but here are the
specifics as I know them to be.

You have four choices and the "don't ask" again option. These options
do the following:

SAVE: Save the file in the current active window and then render
SAVE ALL: Save all open files then render the scene in the current active window
NO: Do not save but render the scene in the current active window
  CANCEL: Cancel the render operation.

Once you select the "do not ask again" option it will act differently.
It will automatically perform the selected task you pick at the time you
chose the "do not ask" option. For example if you choose save then it
saves the current active window each time you hit the render button. I
am not sure what will happen if you choose the cancel option but I
wouldn't try it unless you don't care to render your scene. Once you
have choosen one of the above options there is no way to reset it that
I know of other than closing the program and restarting it.
